An international team of scientists has detected individual hormone and commonly used drug receptors at work in intact cells for the first time using a groundbreaking live-cell microscopy technique.

G protein-coupled receptors (GPCRs) are one of the “hottest” therapeutic targets for diseases like hypertension, asthma, and Parkinson’s disease. Many hormones and neurotransmitters function on these receptors, which enable them to control the behavior of our cells. As a result, these receptors are targeted by about half of all commonly administered medications to treat a variety of diseases.
The use of a highly sophisticated method based on single-molecule microscopy allowed these detailed observations at the cell membrane of living cells. The researchers have discovered that receptors and G proteins are normally only associated for a brief period of time. In reality, most interactions lasted less than a second. Furthermore, the cytoskeleton underneath the cell membrane is implicated in the creation of hot spots, according to the researchers. The presence of these receptor signaling hotspots was previously unknown.
